چکیده

Mastitis is one of the most economic disease affecting dairy cows worldwide. Its classic diagnosis using bacterial culture and biochemical findings is a difficult and prolonged method. In this research, using of matrix-assisted laser desorption/ionization time-offlight mass spectrometry (MALDI-TOF MS) permitted identification of different microorganisms with high accuracy and rapidity (only 24 hours for microbial growth and analysis). During the application of MALDI-TOF MS, one hundred twenty strains of Staphylococcus and Streptococcus species isolated from milk of cows affected by clinical and subclinical mastitis were identified, and the results were compared with those obtained by traditional methods as API and VITEK 2 Systems. 37 of totality 39 strains (~95%) of Staphylococcus aureus (S. aureus) were exactly detected by MALDI TOF MS and then confirmed by a nuc-based PCR technique, whereas accurate identification was observed in 100% (50 isolates) of the coagulase negative staphylococci (CNS) and Streptococcus agalactiae (31 isolates). In brief, our results demonstrated that MALDI-TOF MS is a fast and truthful technique which has the capability to replace conventional identification of several bacterial strains usually isolated in clinical laboratories of microbiology. Keywords—Identification, mastitis pathogens, mass spectral, phenotypical.

Coagulase-negative staphylococci-emerging mastitis pathogens.

Coagulase-negative staphylococci (CNS) have become the most common bovine mastitis isolate in many countries and could therefore be described as emerging mastitis pathogens. The prevalence of CNS mastitis is higher in primiparous cows than in older cows. CNS are not as pathogenic as the other principal mastitis pathogens and infection mostly remains subclinical. However, CNS can cause persisten...

Pathoproof Mastitis Pcr Assay and Sensitivity Validation of a New Test Kit for Rapid Identification of Staphylococcus Aureus, Streptococcus Agalactiae and Mycoplasma Bovis from Bovine Milk

PathoProof Mastitis PCR Assay is a rapid and accurate system for identifying mastitis pathogens from fresh or preserved bovine milk. The system includes: 1) reagent kits; 2) a real-time PCR instrument, i.e. the machine using the reagent kits for analysis of the milk samples; and 3) software for automated interpretation of the results. Rapid and efficient identification of the severe contagious ...

Changing trends in mastitis

The global dairy industry, the predominant pathogens causing mastitis, our understanding of mastitis pathogens and the host response to intramammary infection are changing rapidly. This paper aims to discuss changes in each of these aspects. Globalisation, energy demands, human population growth and climate change all affect the dairy industry. In many western countries, control programs for co...

6 the Mastitis Problem

Mastitis implies that an infectious agent is present in the mammary gland and is nearly always caused by bacteria. Bacteria invade the udder by entering the teat orifice, multiply and die within the gland, and in the process, produce and release toxins that cause injury to secretory tissue and stimulate an immune response. Besides bacteria, other pathogens such as yeast, Mycoplasmas, and algae ...

Invited review: The role of contagious disease in udder health.

Contagious diseases are a threat to animal health and productivity, both nationally and at the farm level. This makes implementation of biosecurity measures to prevent their introduction and spread within countries and farms a necessity. Mastitis is the most common and costly contagious disease affecting dairy farms in the western world. The major mastitis pathogens are endemic in most countrie...
